It depends on the day of the week. On a Saturday or Sunday you would probably be OK. On a Friday the traffic will likely be terrible. I would be safe and take the subway -- you'll make it no problem.
The #4 train and D trains make stops at 161st St./Yankee Stadium. I think your best bet would be to take the D train to the 7th avenue stop. I am pretty sure you'll only be a few blocks from the theatre, but it worth double checking -- just give the theatre a call.
Good luck!

As Genevieve said, the train is fastest and there is a station at either 53rd street or 54th street and 7th Ave that will leave you about two blocks from the theatre. You don't have to worry about being there exactly for three o'clock. I got into see a Letterman taping one day by just walking by the theatre (it was getting fairly close to show taping time) and I was on line for maybe 15 minutes (assistants kindly handed out Snickers bars, lol!) and soon enough I was inside and the taping began! I live in NYC and hadn't planned at all that day to see the Letterman show but a friend I was walking with said to me as we got near the theatre, you should go (he'd gone earlier in the week) and so I did. It was fun. And it's true what they say about the theatre (if you haven't heard, then hear me now!) it REALLY IS COLD IN THERE so make sure you have a sweater or windbreaker in your day pack because once inside, the chill sets in.
Inside, the set looks so much smaller than it does on television, but I guess that's part of the magic of TV! When I was there - I don't know why I remember this - the guests were Bryant Gumbel and the band Everclear. Overall, it was a fun "New York" experience! Especially if the Yankees win the game you'll be attending prior to the Letterman show!!
GO YANKEES!
Enjoy your visit in NYC! We love seeing visitors and helping them out so y'all enjoy your trip and not spend too much time staring at the maps. For example, when you come out from the subway and don't know what direction to walk in order to get where you want to be, just ask one of us and we'll gladly point you in the right direction. Contrary to some folks beliefs, we are not all gun-toting nuts. We just live in a big city, which takes a lot of energy and focus (and crime is down) so if we seem aloof, it's not you, it's us. You asking us a question could be a 'welcome back to earth check' and get us out of our robotic droning tendencies!! It's a fun city but it is BIG so don't hesitate to ask!! Our city belongs to the world so come and have fun!
